Test principle:This assay employs a two-site sandwich ELISA to quantitate LT-B4 in samples. An antibody specific for LT-B4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyLT-B4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for LT-B4 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LT-B4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Leukotriene B4 is a leukotriene involved in inflammation. It is produced from leukocytes in response to inflammatory mediators and is able to induce the adhesion and activation of leukocytes on the endothelium, allowing them to bind to and cross it into the tissue. In neutrophils, it is also a potent chemoattractant, and is able to induce the formation of reactive oxygen species and the release of lysosome enzymes by these cells. Leukotriene B4 is a potent pro-inflammatory molecµLe that belongs to a family of eicosanoid lipid mediators. It is syntheQuantityd from arachidonic acid that is generated from nuclear membrane phospholipid. In general, and upon cell activation, phospholipase A2 is directed to the nuclear membrane. Here, it generates arachidonic acid that is bound by one 24 kDa membrane-bound protein termed Five-Lipooxygenase Activating Protein (FLAP).
Stability:The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
