Test principle:This assay employs a two-site sandwich ELISA to quantitate IL12B in samples. An antibody specific for IL12B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyIL12B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for IL12B is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL12B bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages and B-lymphoblastoid cells (NC-37) in response to antigenic stimµLation. IL-12 is involved in the differentiation of naive T cells into Th0 cells which will further develop into either Th1 cells or Th2 cells. It is known as a T cell stimµLating factor, which can stimµLate the growth and function of T cells. It stimµLates the production of interferon-gamma (IFN-?) and tumor necrosis factor-alpha (TNF-?) from T and natural killer (NK) cells, and reduces IL-4 mediated suppression of IFN-?. T cells which produce IL-12 have a coreceptor, CD30, which is associated with IL-12 activity.IL-12 plays an important role in the activities of natural killer cells and T lymphocytes. IL-12 mediates enhancement of the cytotoxic activity of NK cells and CD8+ cytotoxic T lymphocytes.
Stability:The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calcµLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).
